Analysis of Mitochondrial DNA Mutation in Stomach Cancers

Abstract
Mitochondrial DNA (mtDNA) mutation was tested inhuman stomach cancer using polymerase chain reaction (PCR) and in situ hybridization technique. PCR assay showed that common deletion(4,977 bp) was more pronounced in cancer region than in normal sample of the same patient in most cases (80%). Biotin-labeled probes for D-loop region and for subunit ¥² of cytochrome c oxidase revealed mtDNA mutation in in situ hybridization in 20% of stomach cancer patients (n=40). These results suggest that mtDNA deletion may be involved in tumorigenesis of stomach tissue.
